What is a CEP for tadalafil API?

CEP stands for Certificate of Suitability to the monographs of the European Pharmacopoeia. For a US filing, no. A CEP is not recognised by the FDA as a substitute for the chemistry, manufacturing and controls support a US application requires. An ANDA or NDA referencing an outside API source normally relies on a Type II Drug Master File referenced under a Letter of Authorization. A supplier offering a CEP in place of a DMF has not met a US requirement.

That said, there are four situations where a US-based buyer should actively want a CEP on the table:

  • Dual-market products. If the same tadalafil tablet is going to the US and the EU or UK, you need US-appropriate support and European support from the same API source. Sourcing a supplier who holds both avoids requalifying later.
  • Ex-US markets that recognise the CEP. A number of regulators outside Europe accept or give weight to a CEP. If your distribution map extends beyond North America, it can save a filing.
  • Corroborating evidence. A current CEP tells you an independent European assessor has looked at the manufacturer’s route and specification. That is not a US credential, but it is not nothing when you are comparing shortlisted sites.
  • Supplier maturity signal. Holding and maintaining a CEP means the manufacturer runs a regulatory affairs function capable of tracking monograph revisions and filing variations. Undocumented traders do not.

What a CEP does not cover

  • It is not a GMP certificate. EDQM certification and GMP inspection are separate processes with separate outcomes.
  • It does not cover the finished dosage form. A CEP applies to the active substance, not to your tablet.
  • It does not certify a batch. Batch conformity is evidenced by the Certificate of Analysis, and the two documents answer different questions.
  • It is site-specific and route-specific. Material made at a second site, or by an alternate route, is outside the certificate’s scope even when the company name is the same.

How to verify a tadalafil CEP in five steps

Get the certificate reference and holder name in writing

Every CEP carries a unique reference including a revision indicator. Ask for the full reference and the exact legal name of the holder, not the trading name on the invoice.

Look it up in EDQM’s certification database, not in the PDF

EDQM publishes a searchable database of granted certificates. A PDF proves a certificate existed on the day it was scanned. The database tells you its status today.

Check the revision number and the renewal position

Who should not take tadalafil

  • Anyone with known hypersensitivity to tadalafil or any excipient in the formulation.
